Abstract: | The authors used the reverse hemolytic plaque assay to investigate whether single retinal and optic lobe cells of juvenile and adult crayfish secrete crustacean hyperglycemic hormone (CHH) and whether the secretion rate depends on extracellular serotonin (5-HT) concentration. Nearly 25% of individual retinal and optic lobe cells of juvenile and adult organisms secrete CHH in response to KCl depolarization. In this condition, CHH secretion increased as a function of 5-HT concentration. In both cases, the dose-response curve indicates two different populations of CHH-secreting cells. juveniles showed a higher CHH secretion index than did adult organisms, demonstrating a developmental interstage variation of CHH secretion. The authors conclude that (1) retinal CHH-secreting cells correspond to a population of retinal tapetal cells and (2) optic lobe CHH-secreting cells correspond to two subpopulations of CHH of medulla terminalis-X organ.
